PERBANDINGAN PENGGUNAAN KOH DAN NaOH DALAM DEPROTEINASI SAMPEL SPUTUM
UNTUK ISOLASI DNA MYCOBACTERIUM TUBERCULOSIS

Introduction : Tuberculosis (TB) is an infectious disease of the lungs caused by the bacteria Mycobacterium tuberculosis. According to the WHO in 2010, Indonesia was ranked fifth in the number of patients with pulmonary tuberculosis by 5.8% of the number of people with TB in the world. In general, the diagnosis of this disease microscopic examination, but there are still many weaknesses.

This study pusposed to compare the two compounds have properties capable of dissolving, neutralizing, and mendeproteinasi sputum samples for Mycobacterium tuberculosis DNA isolation compare 1 N NaOH and KOH 0.5 N.

Method : Sputum samples were used as many as 15 samples were examined by health centers and positively diagnosed with TB in smear microscopic examination, then carried the DNA isolation, PCR, and electrophoresis with pre-treatment deproteinasi before using 1 N NaOH and 0.5 N KOH DNA band seen in electrophoresis measured using image J and processed using SPSS.

The conclusions of this study is the result of a sputum sample DNA isolation by running with 0.5 N KOH better than the results of sputum samples isolation by running with 1 N NaOH.
