Introduction : Examination of leukocyte count is used to determine the percentage of the various types of leukocytes. Visual examination of blood smear that has been dyed is still the main way to calculate white blood cells, but the method is now increasingly comprehensive automated inspection diterima.Salah the examination methods hiting leukocyte automatically using laser-based techniques flowcytometry.

Purpose : This study aims to determine the difference leukocyte count results of the manual method and laser-based flowcytometry.

Methods : This study is a research analitik.Data collected in this research is the primary data with the population is patients undergoing routine hematological examination in Hospital Clinical Laboratory Al-Ihsan. The number of samples taken is 50 samples.

Results : From a statistical test to the results of leukocyte count (basophils, eosinophils, neutrophils, lymphocytes and monocytes) p value = 0.000 (< 0.05) in basophils, eosinophils while at p = 0.854 (> 0.05), monocytes p = 0.744 (> 0.05), neutrophil p = 0.744 (> 0.05), and lymphocytes p = 0.310 (> 0.05).

Conclusions : From this study it can be concluded that there are significant differences in the results of basophil counts, but there is no significant difference in the results of the counts of eosinophils, neutrophils, lymphocytes and monocytes using manual methods and laser-based flowcytometry.

Keywords : count of leukocytes, laser-based flowcytometry, manual.
